Effects of re-expressed RASSF1A on gene expression in rhabdoid tumor of the kidney cells.

(Submitter supplied) Several groups reported the correlation between hypermethylation of RASSF1A and aggressiveness or a poor outcome in various pediatric tumors, including Wilms tumor. In addition, it is known that the overexpression of RASSF1A promotes apoptotic cell cycle arrest and reduces the tumorigenicity. In an attempt to identify epigenetic targets related to the biological features of rhabdoid tumor of the kidney (RTK), we analyzed the methylation level of the RASSF1A gene in RTK. more...

Microarray expression data of A375 cells at different density/culture system

(Submitter supplied) Ferroptosis is one of the regulated cell deaths, induced by the accumulation of lipid peroxidation. Induction of ferroptosis is often influenced by the cell environment; however, much remains unknown about cellular states altering ferroptosis susceptibility. We found that melanoma cell lines become resistant to ferroptosis along with an increase in cell density. To elcidate the mechanism, we have investigated the differences in gene expression between A375 cells at different densities/culture systems.

Laser Capture Microdissection Provides a Novel Molecular Profile of Human Primary Cutaneous Melanoma

(Submitter supplied) Melanoma accounts for the majority of skin cancer related mortality. In-depth molecular analysis of neoplastic melanocytes in whole tissue biopsies is diluted by the inflammatory infiltration, which may obscure gene signatures specific to neoplastic cells. Thus, a method is needed to precisely uncover molecular changes specific to tumor cells from a limited sample of primary melanomas.

Agents which upregulate CD55 [decay accelerating factor (DAF)] expression [Array]

(Submitter supplied) (Submitter supplied) Identification of transcription factors (TFs) which upregulate human CD55 expression. CD55 was originally described as a cellular complement regulator that protects self-cells from autologous complement attack. It is now known as cellular regulator which controls many functions of cells, as examples T cell commitment to T effector cells vs Foxp3+ T regulatory cells, B2 cell Ab production, and receptor tyrosine kinase (RTK) growth factor receptor function. more...

Effect of the Foxp3 inhibitory peptide P60 on gene array expression of Karpas cell line

(Submitter supplied) By using a phage displayed peptide library we identified peptide P60 as able to bind Foxp3 and inhibit Treg activity. To gain insight into the effect of P60 in Foxp3+ve cells we used Karpas 299 human cell line derived from a human lymphoma with a regulatory T cell profile. Microarray analysis of Karpas cell line incubated with or without P60 peptide was carried out using the Affymetrix human U133 Plus 2.0 array.

Gene expression data of APC-mutated colorectal cancer patient-derived cells treated with tankyrase inhibitors

(Submitter supplied) Tankyrase, a poly(ADP-ribose) polymerase family member, regulates multiple cellular processes. Tankyrase inhibitors efficiently suppress APC-mutated colorectal cancer (CRC) cell proliferation. To examine the mechanism of anti-proliferative effect of tankyrase inhibitors (G007LK and RK582), we analyzed gene expression profiles of patient-derived APC-mutated CRC cells (JC21 and JC475) left untreated or treated with 0.3 µM G007LK or RK582 for 24 h by cDNA microarray analysis.

Monocytes in Type 1 diabetes families exhibit high cytolytic activity and subset abundances that correlate with clinical progression

(Submitter supplied) Monocytes are immune regulators implicated in the pathogenesis of Type 1 diabetes (T1D), an autoimmune disease that targets the insulin-producing pancreatic β-cell. We determined that monocytes of recent onset (RO) T1D patients and their healthy siblings express proinflammatory/cytolytic transcriptomes and hyper-secrete cytokines in response to lipopolysaccharide exposure compared to unrelated healthy controls (uHC). more...

CD34+ monocyte subset is highly susceptible to HIV-1 infection

(Submitter supplied) HIV-1 persists in cellular reservoirs despite effective antiretroviral therapy (ART). CD4+ T cells are the well-known reservoir, but there is growing evidence that myeloid cells, including circulating monocytes, are also a clinically relevant reservoir. However, it is unclear what are preferentially infected monocyte subsets in vivo. Here, we show that a monocyte fraction expressing the stem cell marker CD34 is more susceptible to HIV-1 infection than the CD34-negative major subset. more...

Expression data from primary human healthy donor-derived CD14- cDC2s, CD14+ cDC2s and monocytes

(Submitter supplied) The human dendritic cell (DC) family has recently been expanded by CD1c+CD14+CD163+ DCs, introduced as DC3. DC3 are found in tumors and peripheral blood of cancer patients but which cells can serve as their pre-cursors remain unknown. CD1c+CD14+ cells share similarities with both CD1c+ DCs (cDC2s) and CD14+ monocytes on transcriptomic and phenotypic level. To investigate whether CD14+ cDC2s are closer related to CD14- cDC2s or monocytes on transcriptomic level, we analyzed their RNA.

Expression data from Lund Human Mesencephalic cells (LUHMES) exposed to acute and persistent oxidative DNA damage

(Submitter supplied) Oxidative DNA damage in neurons activates a DNA damage response (DDR) to promote repair. Under a chronic oxidative environment these processes may be altered and promote accumulation of unrepaired DNA damage and continued activation of a DDR, leading to apoptosis or senescence activation. Accumulation of oxidative DNA damage are features of brain ageing and neurodegeneration but the effects of persistent DNA damage in neurons are not well characterised. more...

Glutamine deficiency drives transforming growth factor-β signaling activation that gives rise to myofibroblastic carcinoma-associated fibroblasts

(Submitter supplied) Tumor-promoting carcinoma-associated fibroblasts (CAFs), abundant in the mammary tumor microenvironment (TME), maintain transforming growth factor-β (TGF-β)-Smad2/3 signaling activation and the myofibroblastic state, the hallmark of activated fibroblasts. How myofibroblastic CAFs (myCAFs) arise in the TME and which epigenetic and metabolic alterations underlie activated fibroblastic phenotypes remain, however, poorly understood. more...

Three-dimensional spherical cellular aggregates of RPTEC as an in vitro tool for evaluation of human renal proximal tubular toxicity and drug disposition

(Submitter supplied) The proximal tubule plays an important role in the secretion and reabsorption of drugs in the kidney and is a major site of drug interaction and toxicity. Kidney toxicity analysis via in vitro assays is challenging as few provide appropriate proximal tubular cell function. In this study, we aimed to develop a simple and reproducible method to culture human proximal tubular epithelial cells (RPTECs), used in pharmacokinetic and toxicological evaluation by monitoring organic anion transporter (OAT)1 as a selection marker. more...

Expression of retinoblatoma

(Submitter supplied) Retinoblastoma is the most frequent intraocular malignancy in children. Little is known on the molecular basis underlying the biological and clinical behavior of this cancer. Loss of KDM6B confers resistance to lipotoxicity through epigenetic downregulation of G0S2 expression in NASH-related HCC

(Submitter supplied) Background& Aims: The incidence of nonhepatitis B and nonhepatitis C viral (NBNC)- HCC has been rising in recent years due to the increase in NAFLD and NASH worldwide with fibrosis. However, the molecular mechanisms underlying the progression of these HCC are not fully understood. Here we observed that KDM6B, an H3K27 demethylase, was commonly downregulated in human NBNC-HCC and mouse NASH-related HCC by microarray analysis. more...
