Symbiont-mediated defense against Legionella pneumophila in amoebae

(Submitter supplied) Legionella pneumophila are important opportunistic pathogens for which environmental reservoirs such as protists are crucial for the infection of humans. Free-living amoebae are considered key hosts providing nutrients and shelter for highly efficient intracellular proliferation of L. pneumophila, which eventually leads to lysis of the amoeba host cell. Yet, the significance of other bacterial players for L. more...

Biphasic metabolism and host interaction of a chlamydial symbiont

(Submitter supplied) Chlamydiae are obligate intracellular bacteria comprising well-known human pathogens and ubiquitous symbionts of protists, which are characterized by a unique developmental cycle. Here we comprehensively analyzed gene expression dynamics of Protochlamydia amoebophila during infection of its Acanthamoeba host by RNA sequencing. This revealed a highly dynamic transcriptional landscape, where major transcriptional shifts are conserved among chlamydial symbionts and pathogens. more...

A systematic spatial bias in microarray hybridizations caused by probe spot position-dependent variability in lateral diffusion

(Submitter supplied) The hybridization of nucleic acid targets with surface-immobilized probes is a widely used assay format for high-throughput detection of many parallel targetsin medical and biological research. Though commonly applied, microarray technology still suffers limitations arising from problems of data robustness and reproducibility across platforms, stemming in part from an incomplete understanding of the complex processes governing surface hybridization behavior. more...

Whole genome oligonucleotide microarray for Protochlamydia amoebophila

(Submitter supplied) 5´-amino modified oligonucleotide probes were printed onto silylated amine-aldehyde glass slides using a BioRobotics MicroGrid 610/TAS system and 16 MicroSpot 2500 split pins. The average spot diameter was about 150 μm and spot centres were 300 μm apart. The relative humidity during spotting was set to 50% within the spotting compartment. Spotted microarrays were stored one day at 100% humidity. Subsequently, slides were washed by soaking them twice in 0.2% SDS for 2 min and twice in double-distilled water for 2 min. more...
