Effect of the res-2 transcription factor gene deletion on protein secretion and stress response in the hyperproducer strain Trichoderma reesei Rut-C30

(Submitter supplied) The fungus Trichoderma reesei is one of the most used industrial cellulase producers due to its high capacity of protein secretion. The strain was shown to possess an expanded endoplasmic reticulum, but the genetic factors responsible for this phenotype remain still unidentified. Recently, three new transcription factors were described in Neurospora crassa which were demonstrated to be involved in protein secretion. more...

Transcriptome study of the deletion of vacuolar protein sorting related vps13 and vps21 in Trichoderma reesei

(Submitter supplied) The disruption of vacuolar protein sorting related genes vps13 and vps21 in filamentou fungi Trichoderma reesei resulted in a dramtic increase in cellulase transcription especially in the late Avicel induction phase, relieving the RESS (REpression under Secretion Stress) effect. Great interest arisen concered about the reason and underlying mechanism of vps13 and vps21 disruption for increased cellulase transcription. more...

The effects of gul1 deletion on the transcriptome of Trichoderma reesei

(Submitter supplied) The morphology of filamentous fungi has a close relationship with the production of many products (e.g. industrial enzymes). In this study, we deleted gul1, which encodes a putative RNA-binding protein, in cellulolytic fungus Trichoderma reesei. The mutant showed different morphologies both on agar plates and in liquid cultures compared with its parent. RNA-seq study showed that the expression levels of genes in many biological processes were affected by gul1 deletion.

Transcriptome analysis of deletion of protein kinase A, catalytic subunit 1 in T. reesei upon growth on cellulose in light and darkness

(Submitter supplied) The aim of this study was to evaluate changes in the transcriptome due to the lack of PKAc1 in light or darkness. As the cAMP pathway represents an output pathway of heterotrimeric G-protein signaling PKAc1 is likely to be important for regulation of enzymes in response to different nutrient conditions. Also, a light dependent effect was found earlier.

Global Transcriptome and Gene Regulatory Network of MAPK Signaling Pathway in Trichoderma reesei During Sugarcane Bagasse Degradation

(Submitter supplied) The filamentous fungus Trichoderma reesei is a saprophyte involved with polysaccharides cell wall depolymerization, being the most important industrial source of cellulases, which have been used for bioethanol production. The regulation of cellulase expression is controlled at the transcriptional level and in a carbon source-dependent manner. However, the signaling pathways and mechanisms involved in regulating the expression of these enzymes in T. more...

ARA1 regulates not only L-arabinose but also D-galactose catabolism in Trichoderma reesei

(Submitter supplied) Trichoderma reesei is used to produce saccharifying enzyme cocktails for biofuels. There is limited understanding of the transcription factors (TFs) that regulate the release and catabolism of L-arabinose and D-galactose, as the main TF XYR1 is only partially involved. The T. reesei ortholog of ARA1 from Pyricularia oryzae that regulates L-arabinose release and catabolism, was deleted and characterized by growth profiling and transcriptomics along with a xyr1 mutant and xyr1/ara1 double mutant. more...

Genome sequencing and transcriptome analysis of Trichoderma reesei QM9978 reveals vib1 to be essential for cellulase induction

(Submitter supplied) The ascomycete Trichoderma reesei is an industrial producer of cellulolytic and hemicellulolytic enzymes and also serves as a model for investigations on these enzymes and their genes. The strain QM9978 has a cellulase negative phenotype and therefore presents a valuable tool for understanding the mechanisms underlying cellulase regulation. Genome-wide RNA-seq analysis during carbon catabolite repression in Trichoderma reesei reveals a multilevel control of cellulase production and action

(Submitter supplied) The ascomycete Trichoderma reesei is one of the most well studied cellulolytic fungi and widely used in the biotechnology industry, as in the production of second-generation bioethanol. Carbon catabolite repression (CCR) mechanism adopted by T. reesei is mediated by the transcription factor CRE1 and consists in the repression of genes related to the production of cellulase when a readily available carbon source is present in the medium. more...
