Summary
This region is located upstream of the Cd8a (CD8a molecule) coding sequence and contains multiple DNase I hypersensitive sites overlapping positive and negative gene regulatory elements. This sequence is important for directing expression of Cd8 antigen in thermocytes and peripheral T cells. It contains an element that binds SATB homeobox 1 and associates with matrix proteins, and may therefore function as a scaffold/matrix attachment region (S/MAR). Association of Ikaros zinc finger protein with this region can suppress position effect variegation of a transgene. A subregion was also identified as a candidate cis-regulatory module (CRM) in developing mouse thymocytes based on a combination of DNase I hypersensitivity and transcription factor binding. That subregion was validated as an enhancer by high-throughput CapStarr-seq reporter assays showing strong activity in P5424 T cells. [provided by RefSeq, Mar 2023]
