Summary
This region contains multiple conserved elements and DNase I hypersensitive sites overlapping with enhancers of the Cd8a (CD8a molecule) gene. Enhancer fragments in this region direct expression to different T cell lineages, including DP thermocytes and mature CD8+ T cells. These sequences may synergize with enhancers located nearby in the CII and CIII DNase I hypersensitive regions. Binding by the POZ (BTB) and AT hook containing zinc finger 1 protein suppresses enhancer activity, causing variegated expression. A subregion was identified as a candidate cis-regulatory module (CRM) in developing mouse thymocytes based on a combination of DNase I hypersensitivity and transcription factor binding. That subregion was validated as an enhancer by high-throughput CapStarr-seq reporter assays showing weak activity in P5424 T cells. [provided by RefSeq, Mar 2023]
