Summary
This region represents the promoter of the mesothelin gene. The proximal promoter and 5' UTR exons are derived from sequence related to SINE and long terminal repeat (LTR) elements. This region contains a short more distal element, known as CanScript, that enhances activity in cancer cells through binding by TEA domain transcription factor 1. Activity of this promoter region may be regulated by DNA cytosine methylation and may be influenced by naturally-occurring polymorphisms in this sequence. A subregion was also shown to be an enhancer by the ChIP-STARR-seq massively parallel reporter assay in naive human embryonic stem cells, where it is marked by the H3K27ac and H3K4me1 histone modifications.
