Summary |
We carry out 5-hydroxymethylation DNA immunoprecipitation (hmeDIP) prior to sequencing Ion Proton P1 to report on the genome-wide 5hmC patterns of hepatic in murine hepatic steatosis.

Overall design |
C56BL/6 mice were fed control (11% calories by saturated fat, Research Diets Inc cat number D12329) or high fat (58% calories by saturated fat Research Diets Inc cat number D12331) diet for 17 weeks to induce obesity and hepatic steatosis. DNA was extracted from fresh frozen liver. We carry out 5-hydroxymethylation DNA immunoprecipitation (hmeDIP) prior to sequencing Ion Proton P1 to report locus specific changes in 5hmC induced by obesity. We demonstrate enrichment of 5hmC within gene bodies of induced genes with reciprocal loss of genic 5hmC in downregulated genes.
