Title Epigenetic profiling of OVA-specific CD8+ TILs from mice treated with control, anti-B7S1, anti-PD-1 or anti-B7S1+anti-PD-1 antibodies
Organism Mus musculus
Experiment type Genome binding/occupancy profiling by high throughput sequencing
Summary Combinational blockade of B7S1 and PD-1 exhibits stronger anti-tumor efficacy than monotherapies. In order to further understand the mechanisms whereby blockade of B7S1 or PD-1 inhibits tumor growth, we conducted ATAC-seq analysis of OVA-specific CD8+ TILs from E.G7-bearing mice treated with control antibodies, anti-B7S1, anti-PD-1 monotherapy or combinational therapy.

Overall design 1 million E.G7 were inoculated subcutaneously into mice on Day 0. On Day 21, OVA-specific CD8+ TILs from E.G7-bearing mice treated with control, anti-B7S1, anti-PD-1 or anti-B7S1+anti-PD-1 antibodies(6 mice per group) were sorted using BD FACSAria Cell Sorter. The collected cells were then subjected to construction of library for ATAC-seq.
