Title RNA-seq transcriptional profiling in primary human hepatocyte(in vitro/in vivo) and Proliferting human hepatocyte(in vitro/in vivo)

Summary The source of human hepatocytes (HHs) is limited because of the shortage of donor organs and the difficulty to expand them in vitro. We developed a defined medium to expand HHs for about 10,000-fold in numbers. The proliferating HHs are bi-phenotypic, showing partially maintained hepatic features and additionally gained expression of progenitor-associated genes. Importantly, proliferating HHs extensively engraft in injured mouse livers at the level comparable to primary HHs. Moreover, these cells are matured upon transplantation in vivo or differentiation in maturation medium in vitro.
 
Overall design During the culture process, transcriptional profiling in PHH and different passages proliHHs were generated by RNA-seq analysis. Transcriptional profiling in PHH and ProliHH-repopulated Fah-/-Rag2-/-IL2rg-/- mouse liver were generated by RNA-seq analysis.
