Overall design |
RELMa-/- mice were infected with 500 L3 of Nippostrongylus brasiliensis worms and sacrificed at day 10 post infection. Lungs were collected, dissociated and sorted for CD11c+ cells. Cells were plated overnight at 37 deg C in 10% serum media. Cells were serum starved in 1% serum media for 2 hour and treated with 500ng/ml recombinant RELMa or PBS control. Cells were lysed with lysis buffer and an volume equivalent to 1000 cells per microliter was frozen at -80 deg C for NanoString analysis. Five microliter of each sample (~5000 cells) was applied to NanoString.
