Summary |
HCI-13 patient-derived xenograft was treated with vehicle or a tissue-selective androgen receptor modulator (SARM), enobosarm. RNA was extracted from the tumors and microarray was performed. Objective: The objective of the study is to determine the effect of an androgen receptor (AR) agonist or a selective AR modulator (SARM) on hormone receptor-positive breast cancer growth in preclinical models and to determine the underlying mechanism of action.

Overall design |
Methods: Patient-derived xenograft (PDX) HCI-13 (a kind gift from Dr. Alana Whelm, University of Oklahoma) was implanted under the mammary fat pad in female NOD SCID Gamma mice. Once the tumors grew to 100-200 mm3, the animals were randomized and treated orally with vehicle or 10 mg/kg enobosarm. Tumor volume was measured thrice weekly. Thirty days after treatment initiation, the animals were sacrificed and the tumors were snap frozen. RNA was extracted from the tumors using Qiagen RNA isolation kit and its quality, integrity, and quantity were checked. The RNA was then subjected to microarray
