Summary |
We developed diffuse glioma cells with H3F3A_K27M mutation.K27M diffuse glioma cells have high migration ability and maintain close contact with neurons with immature neuron-like properties.K27M cells produce neurotrophic factors and lactic acid and affect neurite growth and differentiation/suppression of surrounding neurons. K27M diffuse glioma cells show an invasion type and gene expression patterns similar to those of human glioma.

Overall design |
We prepared immortalized murine glial cells (OS3; established and obtained from C3H male mouse brain) with H3.3-K27M mutation and control glial cells with the empty vector. We harvested three dishes in each cohort (n=3) and extracted RNA from each dishes. Finally, we perfomred RNA microarray analysis.
