Summary |
Glioma is the most common primary malignant brain tumor in adults. Chemo resistance of temozolomide (TMZ), the first-line chemotherapeutic agent, is a major issue in the management of patients with glioma. Alterations of ATRX constitute one of the most prevalent genetic abnormalities in gliomas. Therefore, it is urgent to further elucidate the role of ATRX contributing to TMZ resistance in glioma. We used microarrays to detail the global programme of gene expression underlying cellularisation and identified distinct classes of up-regulated and down-regulated genes after ATRX knockout.

Overall design |
Briefly, LN229R was infected with viruses containing the CAS9 gene and selected with 10 mg/ml puromycin for 7 days. The cells were subsequently infected with virus carrying small guide (sg) RNAs designed for ATRX. The infection proceeded for 24 h and ATRX gene expression and KO was confirmed by western blot. ATRX-NC and ATRX-KO cells were used for mRNA microarray
