Overall design |
NK cells were isolated from peripheral blood by Ficoll-Paque and then a Miltenyi bead kit. They were cultured for seven days in B0 media with 10 ng/mL IL-15 and platebound isotype antibody, anti-NKG2C, or anti-NKG2A. After seven days cells were pelleted and DNA extracted for bisulfite sequencing and analysis by Ilumina Infinium methylationEPIC Beadchips NA10858_batch 2 (control nucleic acid) and NA10858_batch 1 (control nucleic acid) should not have beta values as they are controls, not samples. We did not generate Beta values for samples 1215 (A,B,C) and 1233 (A,B,C) because the raw data did not have good quality control across the three experimental conditions. Therefore we did not use these for analysis for our paper but submitted the raw data for transparency sake.
