Summary |
In patients with endometriosis, the abnormal immune microenvironment of the pelvic cavity is associated with a large number of macrophages, which participate in the formation and development of ectopic lesions. Different polarization states of macrophages play different biological roles. However, the molecular mechanisms of macrophage polarization remains far from being elucidated. We used microarrays to detail the gene expression underlying macropages polarized by rhFGL2 and identified distinct classes of up-regulated and down-regulated genes during this process.

Overall design |
We isolated PBMC from peripheral blood of healthy volunteer via Ficoll density gradient centrifugation, the CD14 MicroBeads were used for the magnetic positive selection of human CD14+ monocytes from PBMC. Monocytes were further differentiated into macrophage (monocyte-derived macrophage, MDM) by incubation with M-CSF for 6 days. The macrophages were used for RNA extraction and hybridization on Affymetrix microarrays.
