Title RNA-seq analysis after depletion of ALDH1A2 by enhancer blocking in Jurkat cells

Summary RNA-seq analysis was performed in a T-cell acute lymphoblastic leukemia cell line (Jurkat) to analyze gene expression changes after ALDH1A2 depletion.
 
Overall design The inducible single-guide RNA mediated dCas9-KRAB system was used to transcriptionally deplete ALDH1A2 gene expression. Jurkat cells expressing the catalytic dead Cas9 fused with KRAB peptide was first established. The cells were then infected with lentivirus encoding an inducible sgRNA targeting the regulatory element of ALDH1A2 gene. Total RNA was harvested 72hrs upon induction of sgRNA and applied for RNA-seq analysis.
