Title Glycine Cleavage System Determines the Fate of Pluripotent Stem Cells via the Regulation of Senescence and Epigenetic Modifications
Organism Mus musculus
Experiment type Expression profiling by high throughput sequencing
Summary To investigate whether embryonic stem cells exhibit a metabolic state different from that of somatic cells, we performed RNA-seq in mouse embryonic fibroblasts (MEFs), induced pluripotent stem cells (iPSCs) and mouse embryonic stem cells (mESCs). To further investigate the effect of Gldc in mESCs, we performed RNA-seq in V6.5 cells with Gldc knockdown.

Overall design RNA-seq for MEF,IPS and V6.5 cells was performed to analyze the gene expression of these lines. mouse ES cell were transfected with Gldc shRNA or non-targeting control shRNA followed by RNA extraction and sequenced on HiSeq X Ten (Mus musculus).
