Title mRNA stability experiments with control or CNOT1-targeting siRNA treatment in HEK293 cells

Summary Determination of mRNA half-life in HEK293 cells with and without the depletion of CNOT1.
 
Overall design HEK293 cells were treated with control or CNOT1-targeting siRNA for 48hrs. Transcription was then inhibited with 1uM triptolide and RNA samples taken at multiple time points (0, 0.5, 1, 2, 4, 8 & 16 hours after transcriptional inhibition). Poly(A) selected RNA was then subject to RNA-seq. This was conducted for three biological replicates.
