Title Transcriptome and epigenomics characteristics of LPS induced CD127 expressing human monocytes

Summary We performed the transcriptomic and epigenetic analysis for both CD127high and CD127low monocytes under LPS stimulation, and RNA-seq and ATAC-seq were performed accordingly for this purpose.
 
Overall design PBMC were isolated from buffy coat of healthy blood donors by Ficoll gradients, and CD14+ monocytes were isolated from PBMC with anti-CD14 magnetic beads (Miltenyi Biotec). CD14+ monocytes were treated with LPS for 6 h and then were collected for FACS-based sorting of CD127high and CD127low monocytes. Sorted cells were either directly used for ATAC-seq or TRIzol lysed for RNA extraction.
