Summary |
Purpose: RNA-seq was used to evaluate the effect of CCT2 knockdown in HNSCC cell lines. Methods: CCT2 was knocked down in SAS cell lines through shRNA and then RNA was extracted from control cells and knockdown cells. Results: Using an optimized data analysis workflow, we identified 205 differentially expressed genes (DEG), of which 141 genes were down-regulated and 64 genes up-regulated, respectively (fold change >2 and adjust p-value < 0.05). RT-qPCR was used to confirm the reliability of RNA-seq data. KEGG and GSEA analysis indicated that PI3K/AKT signaling pathway might be involved in the function of CCT2, which had been proved by western blotting.
