Title mRNA expression profile of regulatory neutrophil induced by sodiumtaurodeoxycholate

Summary Conventional strategies for the development of new drugs have used chemical compounds or large molecules that act as an agonist, antagonist, inhibitor (enzyme or signal transducer), enzyme cofactor or supplement for normal constituents in the body. Recently, the field of cell therapeutics has received much attention. Ex vivo expansion and/or manipulation is essential in cell therapy, which might limit the entrance of pharmaceutical companies into the cell therapy market. In this paper, we show that we can expand a specific subtype of cells, regulatory neutrophils, in vivo by i.v. injection of a compound which stimulate GPCR19 pathway. The in vivo - expanded cells showed therapeutic efficacy in mice with systemic inflammatory diseases, which was not shown by parental cells. The results of this study demonstrate an innovative novel achievement in developing a new drug, using a strategy similar to cell therapy combined with a small molecule.
 
Overall design We injected sodiumtaurodeoxycholate (HY209; or PBS as a negative control for HY209) to C57BL/6 mice intravenously after the inducing sepsis by injecting LPS (or PBS as a negative control for sepsis) i.p.. Two to three biological replicates each containing MACS-soreted Gr1+ cells from 30-40 pooled spleen of C57BL/6 mice were amplified using Ambion Illumina TotalPrep RNA Amplification kit and arrayed on Illumina MouseRef8 v2 chips
