Title Identification of miR-143 mRNA-targets by AGO-RIP in the myeloid cell line K562

Summary MiR-143 is a miRNA commonly found to be downregulated in a variety of cancers. The goal of this study is the identifacation of miR-143 target mRNAs by next generation sequencing (NGS) in myeloid cells after overxpression of miR-143.
 
Overall design We lentiviraly overexpressed miR-143 in the myeloid cell line K562. After selection of positive cells we isolated RNAs associated with the RNA induced silencing complex (RISC) by immunoprecipitation of AGO2. Isolated RNA of 3 independent experiments was subjected to next generation sequencing.
