Title |
Surrounding 4 weeks rep2 |
Sample type |
RNA |
Source name |
Microdissected surrounding cells, infected
Organism |
Mus musculus |
Characteristics |
time: 4 wk strain: C57BL/6 gender: Female age: 6 weeks tissue: surrounding cells outside the region of liver granulomas infection: Schistosoma japonicum
Treatment protocol |
Mice were euthanised and liver tissues were embedded in OCT compound to prepare frozen blocks for laser microdissection.
Growth protocol |
The complete life cycle of schistosoma japonicum was maintained in our laboratory. The cercarial of S. japonicum were shed from Oncomelania hupensis snail to infect the experimental mice.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA from microdissected materials was extracted by a combination use of Trizol (Invitrogen) and RNeasy Mini kit (Qiagen).
Label |
Cy3
Label protocol |
cRNA was synthesised from total RNA with Illumina Total Prep RNA Amplication kit (Ambion).
Hybridization protocol |
Synthesized cRNA was hybridised to Illumina mouse Ref-8 v2 expression beadchips according to manufacturer instructions.
Scan protocol |
Images on the arrays were scanned using Beadstation 500 at green factor level 2.
Description |
Gene expression of the surrounding cells outside the region of granulomas in the liver infected by Schistosoma japonicum at 4 weeks post infection
Data processing |
Feature extraction of the data was generated using GenomeStudio version 1.1.1 and analysed further using GeneSpring GX 11.
