Growth protocol |
S. pneumoniae serotype 2 D39 strain and serotype 3 ATCC6303 strain were grown to mid-logarithmic phase at 37°C using Todd–Hewitt broth (Difco, Detroit, MI) supplemented with yeast extract (0.5%), harvested by centrifugation at 4000rpm for 15 min, and washed twice in isotonic saline. Suspensions of 2x107 (high dose) and 5x104 (low dose) colony forming units (CFU) in 50µL isotonic saline were used as the inocula.
