A pool of RNA from lung from three male mice, raised at Denver altitude (nearly normoxic), with SM22-rtTA x tetO7-BMPR2delx4+ transgene activated for 1 week. These mice have elevated RV pressures, and dysfunctional pulmonary vascular reactivity.
Lung tissues were flash-frozen in liquid nitrogen within minutes of death, and RNA extracted using an RNeasy kit (Qiagen)
First and second strand complimentary DNA was synthesized using standard techniques. Biotin-labeled antisense complimentary RNA was produced by an in vitro transcription reaction.
Mouse Genome 430 2.0 microarrays (Affymetrix, Foster City, CA) were hybridized with 20 �g cRNA. Target hybridization, washing, staining, and scanning probe arrays were done following an Affymetrix GeneChip Expression Analysis Manual.
