Treatment protocol Adenoviruses were added directly to the culture medium and cells were collected 24hr later. For experiments involving siRNA transfetions, cells were transfected with the indicated siRNAs 48hr prior to infection with adenoviruses encoding either GFP ("Ad-GFP") or PGC-1alpha ("Ad-PGC-1alpha").
Growth protocol Human primary hepatocytes were supplied in Hepatocyte medium (BD). HepG2 cells were grown in EMEM supplemented with 10% FBS. C2C12 cells were grown in DMEM with 20% FBS, and differentiated in DMEM containing 2% donor equine serum and 1uM insulin for 7 days.

Extraction protocol Using the Rneasy kit from Qiagen according to manufacturer's protocol
Label biotin
Label protocol Three hundred ng of total RNA was processed for cRNA synthesis with the Ambion WT Expression Kit (PN4411974). Fragmentation and terminal labeling was peformed with the AffymetrixGeneChip WT Terminal Labeling Kit (PN 900671)
 
Hybridization protocol Hybridization was performed at 45 degrees celcius for 17 hours. The arrays were washed and stained in the GeneChip Fluidic Station 450 (Affymetrix) by using Hybridization, Wash and Stain kit (Affymetrix).
Scan protocol The arrays were scanned by the GeneChip Scanner 3000 7G (Affymetrix).
Description Gene expression data from human primary hepatocytes donor 3 infected with Ad-GFP
Data processing The data expressed as CEL files were normalized by the robust multiarray average (RMA) method with the Expression Console software (Affymetrix).
