Human dendritic cells (DC) were generated in vitro from CD14+ monocytes purified from peripheral blood from healthy donors and cultured for six days with 100 ng/mL GM-CSF and 10 ng/mL IL-4. After six days of culture, DC were harvested, washed and cultured in the absence or presence of IL-17A, with or without 2ng/mL IFN-gamma. RNA was extracted from untreated DC at day 0 (DC) or DC cultured for 12 days with IL-17A (DC-17), without or with IFN-g (DC-G17).

Expression values and absent/present/marginal calls were calculated using the GCOS v1.4 software (Affymetrix). Absolute expression transcript levels were normalized for each chip by globally scaling all probe sets to a target signal intensity of 500.
