Growth protocol C57BL/6 mice were injected for 12-14 days with human recombinant Flt3L (Celldex), skin draining LNs (inguinal , axillary, brachial, popliteal and cervical were pooled and DC were preapred by standard collagenase D isolation technique. For some samples mice were treated with 5ug of LPS IV and skin draining LN were harvested 24 hours later. Peritoneal macrophages were isolated from untreated mice
Extracted molecule total RNA
Extraction protocol RNA was extracted with Trizol reagent, followed by clean-up and DNase I treatment with QIAGEN RNeasy mini kit in accordance with the prescribed protocol provided with the kit. Purity analysis was performed by nanodrop and Eukaryote Total RNA Pico Series II (Agilent)
Label biotin
Label protocol RNA was amplified and hybridized on the Illumina MouseRef-8 v2.0 Expression BeadChip
