Treatment protocol The trachea of each animal was exposed under sodium pentobarbital (Dainippon Sumitomo Pharma, Osaka, Japan) anesthesia, and 50 mg/kg of silica in 0.05 ml sterile saline or 0.05 ml sterile saline was instilled intratracheally, respectively. Mice were sacrificed by excessive doses of anesthesia for the collection of lungs on days (D) 0, 3, 7, 14, 28 and 56 after silica instillation (n = 6 for each time point).

Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol FlashTag is ideally suited to label total RNA samples. For new users, 1μg of total RNA is recommended as starting point for labeling.(Genisphere Technical Support: 877.888.3DNA)
 
Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hours at 48°C and 60 rpm on Affymetrix GeneChip miRNA Arrays. GeneChips were washed and stained in the Fluidics Station 450: Fluidics Script, FS450_0003.
Scan protocol GeneChips were scanned using the GeneChip Scanner 3000.
Description miRNA expression data from lung tissue in early fibrosis phase, day 28 after silica exposure.
Data processing Scanned images of microarray chips were analyzed using Expression Console (Affymetrix) with the default settings. Raw data were processed using different algorithms. Detected Above Background algorithm (DABG) and RMA Background Adjustment algorithm were performed in order to remove the background value.
