| Treatment protocol |
CFW Mice aged 10–12 weeks were subjected to myocardium infarction surgery. Briefly, mice were anesthetized (isoflorin), intubated, and ventilated. Left thoracotomy was performed and left anterior descending artery (LAD) permanently ligated with 7-0 silk. Immediately after LAD ligation, AAV9:Luci and AAV9:hYAP were injected into the myocardium seperately. 5 Days after MI and AAV9 injection, heart apexes were collected and placed on ice in the Trizol solution (Invitrogen).

| Growth protocol |
Mice were kept in a standard mouse room. All animal procedures were approved by the Institutional Animal Care and Use Committee.

Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 0.1ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

| Hybridization protocol |
Following fragmentation, the samples were hybridized on Mouse Gene 2.0 ST arrays at 45C with rotation set at 60 RPM for 16 hours using the GeneChip Hybridization Oven 640. The fluidics stations used for washing and staining are the Genechip Fluidics Station 450.

| Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G.

| Data processing |
The software we use is Affy's Expression Console, version 1.3.1.187. We use the software default setting for the RMA analysis. It's RMA background corrected and sketch-quantile normalized.
