Growth protocol |
Peritoneal MF were isolated from naïve euthanized mouse and plated in 24-well TC dishes into complete RPMI media, with 10% FBS and 15% L929 conditional sup containing MCSF o/n to allow MF adherence. Floaters (B cells, T cells etc) were discarded, fresh pre-warmed media was added over MF and cells stimulated with IFNg for 6hrs before RNA collection

Total RNA from stimulated or control peritoneal MF was isolated using Qiagen RNEasy Kit. DNA was removed using Qiagen Dnase protocol.
