Sample GSM1348216 Query DataSets for GSM1348216
Status Public on Mar 14, 2014
Title Osteoblasts_Lrp5_Knockout_Day7_rep4
Sample type RNA

Source name Osteoblasts_Lrp5_Knockout_Day7
Organism Mus musculus
Characteristics strain background: C57Bl/6
genotype/variation: Lrp5-/-
cell type: primary calvarial osteoblasts
time point: day 7 of differentiation
Treatment protocol Osteoblasts were infected with adenovirus expressing Cre-recombinase or Green fluorescent protein 48 hours prior to inducing osteoblast differentiation.
Growth protocol Cultured for 7 days in the presence of 50ug/ml Ascorbic Acid and 10mM Beta-glycerol phosphate to induce osteoblast differentiation.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using the Qiagen Rneasy Mini Kit.
Label Biotin
Label protocol Biotin-labeled cRNA was synthesized by the total prep RNA amplification kit from Ambion (Austin, TX).

Hybridization protocol cRNA was quantified and normalized to 77 ng/μl, and then 850 ng was hybridized to Beadchips.
Scan protocol The hybridized Beadchips were scanned by Illumina BeadScan confocal scanner. cRNA synthesis, hybridization and scanning was performed by the Genome Sciences Laboratory in the Center for Public Health Genomics at the University of Virginia.
Description Lrp5_7_KO_4
Data processing The expression values were log2 transformed and quantile normalized using the LumiR (v.2.14.2) R package. Matrix non-normalized.txt contains Illumina GenomeStudio (framework 2011.1) generated raw intensity values.
