Treatment protocol |
For cardiac differentiation, iPS cells were culture on suspension during 5 days to allow EBs formation. After 5 days, EBs were plated on gelatin coated dishes and treated with 1% of DMSO and/or 100 ng/ml of NRG-1 protein.

Growth protocol |
iPS cells were maintained in serum containing media with feeder cells. Culture media consisted of 15% of serum (Hyclone) and DMEM high glucose (Gibco) supplemented with 1% P/S (Biowhittaker), 1% L-Glu (Gibco), 0.1 mM b-mercaptoehanol (Gibco), 1% non-essential amino acids (Gibco) and LIF Chemicon).

Extraction protocol |
The cells were harvested with TRIzol Reagent (Invitrogen) and the RNA was extracted according to the manufacturer's instructions. As a last step of the extraction procedure, the RNA was purified with the RNeasy Mini-kit (Qiagen, Hilden, Germany).

The sense cDNA was prepared from 300 ng of total RNA using the Ambion® WT Expression Kit. The sense strand cDNA was then fragmented and biotinylated with the Affymetrix GeneChip® WT Terminal Labeling Kit (PN 900671).

Hybridization protocol |
Labeled sense cDNA was hybridized to the Affymetrix MoGene-2_0-st chip according to the the manufacturer protocols and using GeneChip® Hybridization, Wash and Stain Kit.

Scan protocol |
Genechips were scanned with the Affymetrix GeneChip® Scanner 3000.
