Title obese-control_replicate1
Sample type RNA

Channel 1
Source name muscle obese control
Organism Rattus norvegicus
Characteristics strain: Zucker
tissue: skeletal muscle
gender: male
Extracted molecule total RNA
Extraction protocol For miRNA microarray and qRT-PCR analysis total RNA, including small RNAs, was isolated from muscle samples using the Qiagen miRNeasy Mini Kit (Qiagen, Hilden, Germany), according to the manufacturer’s protocol.
Label Hy3
Label protocol 750 ng total RNA from both sample and reference was labeled using the mercury LNA™ microRNA Hi-Power Labeling Kit, Hy3™/Hy5™ (Exiqon, Denmark).

Channel 2
Source name muscle obese control
Organism Rattus norvegicus
Characteristics strain: Zucker
tissue: skeletal muscle
gender: male
Extracted molecule total RNA
Extraction protocol For miRNA microarray and qRT-PCR analysis total RNA, including small RNAs, was isolated from muscle samples using the Qiagen miRNeasy Mini Kit (Qiagen, Hilden, Germany), according to the manufacturer’s protocol.
Label Hy5
Label protocol 750 ng total RNA from both sample and reference was labeled using the mercury LNA™ microRNA Hi-Power Labeling Kit, Hy3™/Hy5™ (Exiqon, Denmark).

Hybridization protocol Labeled RNA samples were mixed pair-wise and hybridized according to the instruction manual using a Tecan HS4800TM hybridization station (Tecan, Austria).
Scan protocol Afterwards, slides were scanned using the Agilent G2565BA Microarray scanner System (Agilent Technologies, Inc., USA).
Description Biological replicate 1 from 6. obese control
sample number in normalized data file: 4
Data processing The image analysis was carried out using the ImaGene® 9 (mercury LNATM microRNA Array Analysis Software, Exiqon, Denmark). The quantified signals were background corrected (Normexp with offset value 10) and normalized using the global Lowess (LOcally WEighted Scatterplot Smoothing) regression algorithm. The file "Normalized_data.txt contains the normalized signal intensities.
