Title |
Streptozotocin-induced diabetic hypothalamus, rep5 |
Sample type |
RNA |
|
|
Source name |
Streptozotocin-induced diabetic hypothalamus
|
Organism |
Mus musculus |
Characteristics |
strain: C57Bl/6 genotype: wildtype gender: male age: 9 weeks old tissue: brain hypothalamus
|
Treatment protocol |
For STZ-induced diabetes experiments, mice were treated with a single intraperitoneal (i.p.) injection (200 µg/g body weight) of STZ (Sigma). Control mice were injected with the same volume of vehicle.
|
Extracted molecule |
total RNA |
Extraction protocol |
RNA was isolated using an RNeasy kit (Qiagen)
|
Label |
biotin
|
Label protocol |
Double-stranded cDNA synthesis was reverse-transcribed from 10 µg of isolated RNA from each hypothalamus by using the SuperScript (Invitrogen) with an oligo(dT) primer containing a T7 RNA polymerase promoter site. Double-stranded cDNA was purified with Phase Lock Gel (Eppendorf). Biotin-labeled cRNA was transcribed by using a BioArray RNA transcript labeling kit (Enzo).

|
|
Hybridization protocol |
A hybridization mixture containing 15 µg of biotinylated cRNA, adjusted for possible carryover of residual total RNA, was prepared and hybridized to the arrays
|
Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip Scanner 3000
|
Data processing |
Intensity values were quantified by using MAS 5.0 software (Affymetrix)
