cRNA amplification and biotin labeling was synthesized from 150 ng total RNA by means of cDNA synthesis followed by an in vitro transcription reaction in the presence of T7 RNA Polymerase and biotinylated nucleotide analog/ribonucleotide mix (3'IVT Express Kit, Affymetrix Inc, Santa Clara, CA)
Biotin-labeled cRNA was cleanup and quantified with magnetic beads and subsequently fragmented.
