Title |
C57BL/6ByJ strain, brain hemisphere, biological rep3 |
Sample type |
RNA |
Source name |
Brain hemisphere of adult male mouse, cerebellum not included
Organism |
Mus musculus |
Characteristics |
Inbred strain: C57BL/6ByJ; Gender: male; Age: 11-week-old (+/- 1 week)
Treatment protocol |
Animals were killed by cervical dislocation, brains were quickly removed, hemisectioned, and alternate left or right hemispheres were collected in RNA-Later. After processing each hemisphere separately, 10 hemispheres were pooled for one high-density oligonucleotide microarray
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Fragmented cRNA probes were prepared from 7 µg of total RNA according to the protocol of Affymetrix using Superscript Choice System (Invitrogen) and a RNA Transcript Labeling Kit (Enzo, Farmingdale, NY).
Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45oC on GeneChip Mouse Genome 430 2.0. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol |
GeneChips were scanned using the GeneChipR Scanner
Description |
Gene expression data from brain hemispheres of experimentally naive male mice
Data processing |
We used .CEL files produced by GCOS for probe (cell) level Robust Multi-Array Average (RMA)-based normalization of data as implemented in GeneTraffic ver. 3.0
