Title |
TGF-beta24h_SP600125 |
Sample type |
RNA |
Source name |
A20 cells stimulated with TGF-beta in the presence of SP600125 for 24 h
Organism |
Mus musculus |
Characteristics |
Cell line: BALB/c mouse B lymphoma
Treatment protocol |
A20 cells were left untreated or treated for 12 h or 24 h with TGF-beta in the absence or presence of SP600125
Growth protocol |
A20 cells were cultured in RPMI medium supplemented with 10 % fetal bovine serum, 20 mM Hepes, 50 microM beta-mercaptoethanol, 50 units/ml penicillin and 50 microg/ml streptomycin at 37C.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was prepared using the RNeasy Mini Kit according to the manufacturer's instructions (Qiagen).
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 10 microg total RNA.
Hybridization protocol |
Following fragmentation, 15 microg of cRNA were hybridized for 16 hr at 45C on the Affymetrix Mouse Genome 430 2.0 array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol |
GeneChips were scanned using an Affymetrix GeneChip Scanner.
Description |
Gene expression data from A20 cells stimulated with TGF-beta in the presence of SP600125 for 24 h
Data processing |
The data were analyzed with GeneChip Operating Software (GCOS v.1.2) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 500.
