Title |
rat at C3, biological rep2 |
Sample type |
RNA |
Source name |
Rats which were supplied with tap water for 2 weeks.
Organism |
Rattus norvegicus |
Characteristics |
tissue: kidney
Treatment protocol |
experimental (T) group for the gene and protein expression studies and control (C) group. Rats in T group were provided free access to drinking water containing 1% (vol/vol) ethylene glycol and 0.5% ammonium chloride for a period of 2 weeks in order to induce hyperoxaluria, while the rats in C group were supplied with tap water for 2 weeks.
Growth protocol |
all rats were acclimated at 23℃ on a 12 h light/dark cycle before initiation of experiments in the SPF level animal house.
Extracted molecule |
total RNA |
Extraction protocol |
total RNA was performed according to the manufacturer's instructions.
Label |
biotin
Label protocol |
About 1ug total RNA were polyA tailed and labled with FlashTag™ Biotin HSR Labeling Kit(Affymetrix, Santa Clara, CA, USA).
Hybridization protocol |
The labled RNA were hybridized for 16 hr at 60 rpm at 48C on Affymetrix miRNA4.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G
Description |
Gene expression data from rats which were supplied with tap water for 2 weeks.
Data processing |
Raw data files were analyzed by using Affymetrix Expression Console with the default miRNA array settings. Raw data were processed and normalized on basis of organism rat using RMA-DABG algorithm.
