Treatment protocol Embryos at 12.5 dpc were collected and primary embryonic fibroblasts were isolated from them.
Growth protocol Cultured in Dulbecco’s modified Eagle’s medium (DMEM) with 10% fetal calf serum (FCS), penicillin and streptomycin at 37 degree with 5% CO2.
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 2ug total RNA.
 
Hybridization protocol Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45 degree on GeneChip Mouse Genome 430 2.0 Array . GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the GeneChip Scanner 3000.
Description Cas exon 2+/∆ mice were intercrossed and embryos at 12.5 dpc were collected. Heads and inner organ tissues were used for genotyping and primary embryonic fibroblasts were isolated from the remaining of embryos and cultured
Data processing The data were analyzed using GeneSpring (Agilent Technologies).
