Sample GSM213397 Query DataSets for GSM213397
Status Public on Jul 31, 2007
Title Cervical carcinoma 24 Pre-treatment
Sample type RNA

Source name Cervical carcinoma Punch biopsy before hydralazine and magnesium valproate treatment
Organism Homo sapiens
Characteristics Gender: Female
Tumor Stage: Cervical carcinoma IIIB (FIGO)
Treatment protocol Patients received a single oral dose of 500 mg of sulphamethazine early in the morning, and urine was collected within the ensuing 6 h for phenotyping of acetylator status as reported. Afterward, patients began treatment (day –7) with a daily dose of a slow-release formulation of hydralazine tablets containing either 182 mg for rapid, or 83 mg for slow acetylators. Magnesium valproate tablets of 700 mg were also administered as a slow-release formulation at a dose of 30 mg/kg t.i.d. Both were administered until intracavitary therapy was completed. Punch biopsies were taken from primary cervical tumors at diagnosis and at day 8 of treatment with hydralazine and magnesium valproate prior to the first dose of cisplatin and external radiation. Part of the biopsy was sent to the Institution’s Pathology Department for routine hematoxilin and eosin evaluation. The remaining biopsy specimen was immediately frozen at –70°C for biological analyses.
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions
Label Biotin
Label protocol Biotinylated cRNA was prepared according to the standard CodeLink protocol from 2 ug total RNA

Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 37C on Human Whole Genome CodeLink Bioarray. Stain and post-hybridization washes were performed according to the manufacturer's instructions.
Scan protocol Bioarrays were scanned with a GenePix Personal 4100A scanner.
Description None
Data processing Expression values were globally normalized to the median expression value of the whole array spots. To circumvent division by zero, values below three times background value were taken as 5.
