Treatment protocol Oral administration of tamoxifen to wild type and calreticulin transgenic mice to 8-10 weeks-old-mice for 3 weeks was conducted to induce calreticulin expresssion in the heart
Growth protocol Wild type and transgenic mice were grown in standard animal facility with 12 hour light/dark cycle, and 8-10 weeks-old-male-mice were used in this study
Extracted molecule total RNA
Extraction protocol Total RNA was isolated from wild type and transgenic heart homogenates using TRIzol reagent (Invitrogen), and a double strand cDNA library was constructed from 300 ng of purified RNA using a library synthesis kit (Sigma)
Label Cy3
Label protocol Purified cDNA was mixed with Cy3 primer and dNTP/Klenow fragment (50 U/ul) (NimbleGen one color labelling kit), and incubated for 2 h at 37oC with a heated lid
 
Hybridization protocol Hybridization to mouse MM9 Expression Array (12 x 135K, NimbleGen) was carried out at 42oC for 20 h in an HX12 Mixer (Roche), to acquire uniform signal and high sensitivity with minimal sample volume across the array surface
Scan protocol Arrays were scanned using NimbleGen MS 200 software, and the data were extracted using NimbleScan software
Description This sample is of calreticulin transgenic mouse C57BL/6. It is the third of three calreticulin transgenic biological replicates used in this experiment
Data processing Array normalization was performed using the quantile normalization method of Bolstad et al (Bolstad, Irizarry et al., 2003). n-fold change ratios for a particular gene in calreticulin transgenic and control heart were calculated using the RMA-processed expression values (RMA calls)
