Treatment protocol Rats were treated with saline or heroin (1 mg/kg, i.p., b.i.d.) for 7 days. The nucleus accumbens of three rats from each group were mixed and homogenized in TRIzol.
Growth protocol Six-week-old Male Sprague-Dawley rats were purchased from the Shanghai SLAC Laboratory Animal Co. Ltd. and housed with a 12 h reverse dark/light cycle at 23℃. All subjects were allowed free access to food and water. 8-10 weeks old rats were used for experiments.
Extracted molecule total RNA
Extraction protocol Total RNA are harvested using TRIzol (Invitrogen) and RNeasy mini kit (QIAGEN) according to manufacturer’s instructions.
Label Hy3
Label protocol The samples are labeled using the miRCURY™ Hy3™/Hy5™ Power labeling kit.
 
Hybridization protocol The samples were hybridized on a hybridization station. Array = miRCURY LNA microRNA Array, v.11.0 - hsa, mmu & rno. The miRNA associated with the probe ID is available in the .gal file on the series record.
Scan protocol Scanning is performed with the Axon GenePix 4000B microarray scanner. GenePix pro V6.0 is used to read the raw intensity of the image.

Rats were treated with heroin (1 mg/kg, i.p., b.i.d.) for 7 days. The nucleus accumbens of three rats were mixed and homogenized in TRIzol.
Data processing The intensity of green signal is calculated after background subtraction and replicated spots on the same slide have been averaged by getting a median intensity. We use Median Normalization Method to obtain “Normalized Data”, Normalized Data=(Foreground-Background)/median, the median is 50 percent quantile of microRNA intensity which is larger than 50 in all samples after background correction.
The processed data file includes probe ID and median-normalized signal.
