Extraction protocol |
Peripheral blood was collected from each subject at the beginning of the workday around 0900 hr and processed within 6 hr of collection. PBMC were isolated by Ficoll-Paque and lysed in RLT buffer (Qiagen). Lysates were frozen at -80°C and later thawed and extracted using Qiagen's RNeasy mini kit according to the manufacturer's instructions.

cDNA was generated from total RNA (200ng) and used as a template for the in vitro transcription of biotin-labeled cRNA using the Illumina® RNA Amplification kit (Ambion, Austin, TX) according to the manufacturer’s protocol.

Hybridization protocol |
Following fragmentation, 850ng of cRNA were hybridized for 16 hr at 55C on Illumina Sentrix HumanRef-8 chips. Hybridization, washing and detection were performed using Illumina's Gene Expression System Buffer Kit according to the manufacturer's instructions.

Scanning was performed using a BeadArray Reader and BeadScan software (Illumina)
