Treatment protocol C2C12 cells were treated with DMSO (control) or 5uM reversine.
Growth protocol The C2C12 cell line, which is mouse myoblast, was cultured in Dulbecco's modified Eagle's medium (DMEM), supplemented with 10% fetal bovine serum and 1% penicillin/streptomycin.
Extracted molecule total RNA
Extraction protocol Total RNA was isolated using easy-Blue reagent (iNtRON Biotechnology, Sungnam, Gyeonggi, Korea). RNA was stored at -70C in Rnase free H20 supplemented with the Rnase inhibitor. Quality and quantity of RNA was determined using a Agilent Bioanalyzer as per manufacturer procedures(Agilent Technologies, Palo Alto, CA)
Label DIG-UTP
Label protocol 1 ug of total RNA was used to transcribe DIG-labeled cRNA using Applied Biosystems Chemiluminescent RT-IVT kit V2.0.
 
Hybridization protocol Microarray hybridization (using twenty micrograms of fragmented, DIG-labeled cRNA) and processing were performed according to Applied Biosystems protocols.
Scan protocol Chemiluminescence detection, imageing, auto gridding and image analysis was done according to Applied Biosystems protocols and the 1700 Chemiluminescent Microarray Analyzer Software v. 1. 0. 3.
Description C2C12 cells were treated with 5mM reversine. for 4 days.Microarrays were performed in duplicated, R1 and R2
Data processing Signal intensities across microarrays were normalized using the quantile-quantile method (www.bioconductor.org).
