Treatment protocol Keratinocytes were treated with 20ng/ml IL-36α, IL-36β, IL-36γ (Bio-Techne) or vehicle control. Cells were harvested after 24h.
Growth protocol Primary keratinocyte cultures were established as described elsewhere (Mahil et al, J Invest Dermatol 2016) using discarded healthy skin from a donor undergoing a plastic surgery procedure at Guy’s and St Thomas’ NHS Foundation Trust. Keratinocytes were cultured in Epilife medium containing Supplement 7 and 1% penicillin-streptomycin (all reagents Gibco).

Extraction protocol Total RNA was isolated using RNeasy Plus Mini Kit (Qiagen)
RNAseq libraries were prepared with the Illumina TruSeq Stranded Total RNA preparation kit and sequenced on an Illumina HiSeq3000 system.
