Characteristics rats from five experimental groups were sacrificed 19–21 days after surgery
Treatment protocol Rats having the L5-L6 spinal nerves ligated (SNL) were performed, Sham-operation consisted of exposing the spinal nerves without ligation or manipulation. Naïve animals did not receive surgery.Rats were tested before and 12–14 days after surgery. Only SNL rats exhibiting a significantly reduced PWT in the ipsilateral limb were included in drug testing or gene expression studies (eight to twelve rats per group). Vehicle (0.9% saline) or gabapentin (GPN; 100 mg/kg dissolved in 0.9% saline) was administered intraperitoneally (i.p.) once a day for 7 days in a volume of 2 ml/kg. On the last (7th) treatment day (corresponding to 19–21 days post-surgery), rats were again assessed by PWT, in particular to confirm the reversal of neuropathic pain with GPN treatment.

Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol (Expression Analysis Technical Manual, 2001, Affymetrix).
 
Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.

Data processing The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method.
