Extraction protocol |
miRNA was extracted by Assurgen using a proprietary method. cDNA was synthesized with Taqman miRNA Reverse Transcription kit (Thermofisher Scientific) in conjunction with Megaplex RT primers (Pools A & B, Cat #4399966 & # 4444281). cDNA targets were further pre-amplified using Megaplex Preamp Primers (Pool A, Cat # 4399233 & Pool B, Cat # 4444303) & mixed with Taqman OA Real-time PCR Master Mix following the manufacturer's protocol and loaded onto OpenArray Human miRNA plates (Cat # 4461104) using an AccuFill (Applied Biosystems). miRNA panels were run on a QuantStudio 12K Flex instrument.
