1 x 10^4 TIME human MECs were co-cultured with either 2 x 10^4 primary ovarian CAFs or 2 x 10^4 normal ovarian fibroblasts (NFs) in Boyden chambers for 48 hours

Growth protocol |
Ovarian fibroblasts was cultured with medium M105/199 supplemented with 10% Fetal bovine serum and 1ng/mL Epidermal growth factor. TIME cells were cultured in endothelial cell growth medium-2 (Lonza). All cell types were cultured in a humidified incubator at 37C with 5% carbon dioxide.

Total RNA extraction was performed using the Purelink RNA mini kit (Invitrogen) according to the manufacturer's instructions.
