Treatment protocol |
Mature M2 macrophages were transfected with either mimic or inhibitor using the Lipofectamin RNAiMAX reagent according to manufacturer instructions.

Growth protocol |
Blood was obtained from healthy donors. PBMCs from buffy coats were isolated. Subsequently, PBMCs underwent negative selection for CD14 Monocytes. Selected monocytes were then seeded in M2-activation medium . After 6 days additional boost of fresh media were added together with IL-4 (50ng/ml) . After 3 more days (day 9) media were replaced and cytokines were added as in day 6.

Extraction protocol |
RNA was isolated using the miRNeasy Micro Kit (QIagen) according to manufactures protocol. RNA was quantified using a Nanodrop ND-1000 (Thermo Fischer Scientific) and Bioanalyser (Agilent).

Sense strand cDNA was fragmented and labeled using Affymetrix WT terminal labeling kit.

Hybridization protocol |
Four replicates of each group were hybridized to Affymetrix mouse (human) Gene ST 1.0 GeneChip in Affymetrix hybridization oven at 45C, 60rpm for 16 hrs.

Scan protocol |
Wash and stain were performed on Affymetrix Fluidics Station 450 and scanned on Affymetrix GeneChip scanner 3000.
